The present invention relates to a composition comprising recombinant human iduronate-2-sulfatase, a formulation, and a production method thereof. Iduronate 2-sulfatase (IDS) of the present invention has a saccharification pattern and a formylglycine content which are different from a previously known Elaprase, and is also excellent in an enzymatic activity and stability. Thus, the IDS can be effectively used in treatment of Hunter syndrome.
