摘要 Recombinant scFv-immunotoxins target tumor cells expressing human podoplanin but not podoplanin-negative or normal cells. The immunotoxins can be used for treatment of malignant glioma patients or any malignant tumor expressing podoplanin. One such immunotoxin comprises a modified Pseudomonas exotoxin (PE38) attached to the scFv antibody fragment. This immunotoxin can be used as a therapeutic drug for the treatment of malignant gliomas and other cancers.

An immunotoxin which consists of a single polypeptide that binds to podoplanin and which is cytotoxic to cells expressing podoplanin, comprising: an antibody heavy chain variable (“VH”) region and an antibody light chain variable (“VL”) region, each region comprising three complementarity determining regions (“CDRs”), wherein the CDRs of each region are numbered sequentially CDR1 to CDR3 starting from the amino terminus wherein CDR1, CDR2, and CDR3 of the VH are shown in SEQ ID NO: 6, 7, and 8 respectively, and wherein CDR1, CDR2, and CDR3 of the VL are as shown in SEQ ID NO: 9, 10 and 11; and a Pseudomonas exotoxin or cytotoxic fragment thereof (“PE”).
