<p>In order to measure the arrangement structure of water molecules at a specimen-culture fluid interface in a culture fluid, the irregularity of the surface of a specimen in the culture fluid, potential distribution, and composition distributions, arrangement structures, and the like of molecules, protein, and the like with high spatial resolution, a scanning probe microscope is provided with a probe (1), a specimen holder (11) in which a specimen (3) is mounted, an oscillator (2) which displaces the position of the probe, pulse laser beams (22, 23) which are incident on a region measured by the probe in the specimen, a filter-equipped detector (25) which measures the intensity of output light (24) outputted from the specimen by energy spectroscopy, a scanning mechanism (31) which moves the specimen holder, and a control device (26), and measures the shape of the surface of the specimen, and linear/nonlinear optical characteristics. The probe enhanced detection efficiency is optimized by periodically displacing the position of the probe of the oscillator and maximizing the output light intensity dependent on the distance between the probe and the specimen by the control device.</p> |
