摘要 The purpose of the present invention is to eliminate extra-focal fluorescence from focal fluorescence and acquire a sharp fluorescent image. Provided is a microscope (1) that is equipped with: a scanner (7) for scanning excitation light from a light source (2); an objective optical system (8) for focusing scanned excitation light onto a test sample (A), while focusing fluorescence generated in the test sample (A) at each scanned location; a detector (12) for detecting the focused fluorescence; a light shield member (11) situated between the detector (12) and the objective optical system (8), and for selectively blocking the fluorescence focused by the objective optical system (8); a switching unit (3) for switching the positional relationship of the light shield member (11) and the focal point of excitation light on the test sample (A), between an optically coupled positional relationship in which focal fluorescence emitted from the focal point passes through the light shield member (11), and a non-coupled positional relationship in which focal fluorescence is blocked by the light shield member (11); and a computing unit (5) for computing the difference of the fluorescence signals acquired by the detector (12) at the two different positional relationships.
