PROBLEM TO BE SOLVED: To provide an efficient and simple preparation method of recombinant humanμ-calpain.SOLUTION: A preparation method of recombinant humanμ-calpain according to the invention comprises incubating, under a cold condition, a transformation host cell obtained by introducing with both a gene encoding an active protein subunit of humanμ-calpain and a gene encoding a modified control protein subunit with deletion of almost all of GR domain of the control protein subunit of humanμ-calpain so as to enable expression in a host cell; and collecting the recombinant humanμ-calpain from the obtained culture.
