Provided is a Rebaudioside M enzymatic preparation method, the method using Rebaudioside A or Rebaudioside D as a substrate, and in the presence of sucrose and UDP use a mixture of UDP-glucosyltransferase and sucrose synthetase or a recombined cell containing UDP- glucosyltransferase and sucrose synthetase to conduct catalysis, producing Rebaudioside M in the reaction. The reaction is conducted in an aqueous phase system of pH5.0-9.0 at 20-60°C, and the reaction system further contains methylsulfoxide of 3%-5% volume ratio concentration for facilitating substrate dissolving.
