The present invention relates to biosynthesis of polyunsaturated fatty acid and, more specifically, makes a multiple expression vector to co-express three genes of McD6DES, AsELOVL5, and PtD5DES using a pAO815 vector and transforms the vector to Pichia pastoris which is yeast using methane being a usable system used in producing protein of an eukaryotic cell, thereby valuably being used as a polyunsaturated fatty acid producing source because of analyzing transformation from an extracellular matrix produced by the yeast, such as linoleic acid (LA, C18:2 n-6) or a-linolenic acid (ALA, C18:3 n-3) to the polyunsaturated fatty acid such as gamma-linolenic acid (GLA, C18:3 n-6), stearidonic acid (STA, C18:4 n-3), di-homo-linolenic acid (DGLA, 20:3n-6), eicosatetraenoic acid (ETA, 20:4n-3), arachidonic acid (ARA; 20:4n-6), eicosapentaenoic acid (EPA, 20:5n-3), docosatetraenoic acid (DTA, 22:4n-6) or docosapentaenoic acid (DPA, 222:5n-3) by using gas chromatography to check the co-expression function of the three genes. |
