A method of treating a disease or condition in which up-regulating GAGs is therapeutically beneficial is disclosed, in particular osteoarthritis and skin diseases. The method comprises locally administering to a subject a therapeutically effective amount of an agent capable of down-regulating activity or expression of a component of the renin-angiotensin system.

A method of treating osteoarthritis in a subject, the method comprising intra-articularly administering to a subject in need thereof a therapeutically effective amount of an agent capable of down-regulating activity or expression of a component of the renin-angiotensin system, wherein said agent is selected from the group consisting of an angiotensin converting enzyme (ACE) inhibitor, an AT1 receptor antagonist and a renin inhibitor, thereby treating osteoarthritis in the subject.
