PROBLEM TO BE SOLVED: To provide a method for recombining nucleic acids including homologous nucleic acids, to provide a family of gene shuffling oligonucleotides and its use in recombination procedures, and to provide a polymerase- and ligase-mediated recombination method.SOLUTION: There is provided the method for oligonucleotide-assisted shuffling of nucleic acids. The oligonucleotide-assisted approaches especially facilitate a family shuffling procedure and provide a substantially simplified shuffling protocol. The protocol can be used for generating a family-shuffled nucleic acid without isolating or cloning whole-length homologous nucleic acids.
