METHODS OF MC4R GENOTYPES ON CARCASS TRAITS IN HANWOO POPULATION

PURPOSE: A method for improving carcass traits of Hanwoo(Korean beef) using genetic polymorphism of melanocortin 4 receptor(MC4R) gene is provided to improve economic traits of Hanwoo using genotype. CONSTITUTION: A primer is described on tablet for analyzing genotype of C927T, C1069G, C1343A, and C1786T of MC4R gene in Hanwoo. DNA is isolated from a tissue sample using QuickGene DNA tissue kits(FUJIFILM). The extracted genomic DNA is used in a test by measuring concentration and purity with ND-1000 UV-Vis Spectrophotometer(NanoDrop Technologies, USA). Other bands are removed from the amplified product of MC4R gene through purification. An individual base sequence for each primer is determined by adhering a fluorescent dye to each base of the amplified product using BigDye Terminator v3.1 Cycle Sequencing Kits(Applied Biosystems, USA). The mutation of the determined sequence is identified using seqMAN II(DAN ATAR Inc.) program.
