588112 EXPRESSION-LINKED GENE DISCOVERY Provided is an in vitro method for the identification of expressed genomic DNA in a sample, comprising isolation of mRNA from a sample of a selected organism and preparing from said mRNA as a template small single stranded cDNA fragments with one adaptor containing an affinity label; isolation of genomic DNA from a sample of the same or a related organism and preparing from said genomic DNA single stranded genomic DNA fragments ligated to adaptor molecules; hybridization of said single stranded genomic DNA fragments with said single stranded cDNA fragments and amplification of said hybrids; and high throughput sequencing of said hybrids.
