METHOD FOR HIGHLY EXPRESSING TARGET GENE BY PUTTING AMPLIFICATION STRUCTURE OF THE SAME UNDER CONTROL IN MAMMALIAN CELL

PROBLEM TO BE SOLVED: To provide a method for highly expressing a target gene by making hard to cause the expression inhibition of the target gene stemmed from an inversion repetitive sequence developed in a gene amplification method.

SOLUTION: The method for highly expressing a target gene in a mammalian cell is provided, comprising a linking step of making a serial repetitive sequence by consecutively linking a plurality of straight-chain polynucleotides each with both terminal ends representing non-palindrome-projected ones, each containing a gene expression cassette containing the target gene linked controllably to a promoter; a gene-transferring step of transferring the thus obtained serial repetitive sequence into the mammalian cell; and a step of expressing the target gene in the resultant transformed cell obtained by the preceding step.
