Described is a method, oligos and kit for detecting the phytoplasma pathogen microorganism causing the lethal yellowing disease in plants, which comprises the steps (a) of obtaining a biological sample of the plant; (b) isolating the DNA of the sample plant, (c) amplifying the same by PCR in real time with an oligonucleotide set which comprises, a sense oligonucleotide comprising the sequence 5'-GCTAAAGTCCCCACCATAACGT-3', and an antisense oligonucleotide comprising the sequence 5'-CGTGTCGTGAGATGTTAGGTTAAGT-3', and a probe that consists in a sequence 5'-CCCCTGTCGTTAATTG-3'; (d) detecting the fluorescence emitted by the probe during the amplification method by PCR in real time, and (e) determining the presence of the phytoplsama pathogen microorganism causing the lethal yellowing in plants, where the amount of fluorescence is proportional to the amount of DNA in the phytoplsma pathogen microorganism causing the lethal yellowing in plants.
