<P>PROBLEM TO BE SOLVED: To provide an amino acid sequence and a nucleotide sequence of a humanized monoclonal antibody, and its use. <P>SOLUTION: There is provided the humanized monoclonal antibody binding to tumor necrosis factor α. The humanized monoclonal antibody contains a light chain variable region containing a specific amino acid sequence and a heavy chain variable region containing a specific amino acid sequence, wherein the amino acid sequence contains at least one amino acid substitution, and the amino acid substitution comprises at least one substitution selected from the group consisting of substitution of position 10 amino acid isoleucine in the light chain variable region with threonine, substitution of position 18 amino acid lysine with arginine, substitution of position 2 amino acid lysine in the heavy chain variable region sequence with glutamine, substitution of position 10 amino acid tryptophan with leucine, substitution of position 18 amino acid lysine with arginine and substitution of position 41 amino acid glutamic acid with glycine. <P>COPYRIGHT: (C)2012,JPO&INPIT |
