Provided is a method for generating autologous pancreatic stem cells and autologous islets through reverse differentiation of human somatic cells. The method comprises culturing human somatic cells stepwise by using a culture media containing different plant extracts and protein ingredients, so that the somatic cells are reversely differentiated to generate autologous pancreatic stem cells, which are further cultured to generate islets. Through the method, tens of billions of passage 1 pancreatic stem cells and autologous islet cells are generated in 2-3 weeks, which are suitable for establishing a new autologous pancreatic stem cell bank for permanently depositing the autologous pancreatic stem cells. The plant and protein induced human autologous pancreatic stem cells have a cell-specific phenotype and pluripotency in cell differentiation similar to those of the human natural pancreatic stem cells. The autologous pancreatic stem cells and the autologous islet cells are not only applicable in an application field of pancreatic diseases comprising insulin-deficient diabetes, but also have a great potential in the field of autologous pancreas tissue engineering.
