Production of (1S,4R)-1-amino-4-hydroxymethyl-2-cyclopentene (Ia) or (1R,4S)-1-amino-4-hydroxymethyl-2-cyclopentene (Ib) or their N- and/or O-acylated derivatives comprising optical resolution of racemic 1-amino-4-hydroxymethyl-2-cyclopentene (II) either by reaction with optically active tartaric acid or by treatment with a hydrolase in the presence of an acylating agent, is new. Independent claims are also included for: (1) producing (Ia) or (Ib) by chemical hydrolysis of their N- and/or O-acylated derivatives; (2) producing (1S,4R)- or (1R,4S)-4-(2-amino-6-chloro-9H-purin-9-yl)-2-cyclopentene-1-methanol by reacting (1S,4R)- or (1R,4S)-1-amino-4-hydroxymethyl-2-cyclopentene hydrogen D- or L-tartrate with N-(2-amino-4,6-dichloro-5-pyrimidinyl)formamide and cyclizing the resulting (1S,4R)- or (1R,4S)-4-(2-amino-6-chloro-5-formamido-4-pyrimidinyl)-2-cyclopentene-1-methanol; (3) (1S,4R)- or (1R,4S)-1-amino-4-hydroxymethyl-2-cyclopentene hydrogen D- or L-tartrate. |
