PROBLEM TO BE SOLVED: To provide a method of making it possible to produce a porous membrane with an affinity mechanism which is suitable for separation and purification of protein during a simple step.SOLUTION: The porous membrane includes: a polymer; an organic liquid with affinity to polymer; and an organic compound which has an affinity ligand and a hydrophobic chain combined with the affinity ligand. The method includes a step of forming a phase separation mixture which is layer-separated into a dense phase with high concentration of the polymer and a dilute phase with the concentration of the polymer lower than the dense phase.
