PROBLEM TO BE SOLVED: To provide a practical method for amplifying a DNA.SOLUTION: The method for amplifying the DNA includes treating the DNA with one or more kinds of restriction enzymes forming an overhang having a length of five or more bases and containing one or more arbitrary base sites when amplifying the DNA to prepare a first double-stranded DNA fragment, linking to the first double-stranded DNA fragment, an adaptor DNA having a linkage sequence containing a base sequence complementary to the base sequence of the overhang and an amplification sequence containing a common base sequence for the amplification to acquire a second double-stranded DNA fragment, and amplifying the second double-stranded DNA fragment by using a DNA polymerase. |
