MODIFIED DOUBLE-STRANDED RNA

Modified double-stranded RNA having the following characteristic properties (1) to (5): (1) the RNA has a sense strand which comprises a target nucleotide sequence that is complementary to a nucleotide sequence for mRNA of a target gene and has a length of 30 to 100 nucleotides; (2) the RNA has an antisense strand which comprises a nucleotide sequence complementary to the target nucleotide sequence: (3) at least two contiguous nucleotides located in a region lying between position-22 to position-24 from the 5'-terminal of the sense strand and/or at least two contiguous nucleotides located in a region lying between position-22 to position-24 from the 3'-terminal of the antisense strand are nucleotides in each of which a hydroxy group at position-2' in a ribose is substituted; (4) at least two contiguous nucleotides located in a region lying between position-22 to position-24 from the 3'-terminal of the sense strand and/or at least two contiguous nucleotides located in a region lying between position-22 to position-24 from the 5'-terminal of the antisense strand are nucleotides in each of which a hydroxy group at position-2' in a ribose is substituted; and (5) the RNA has an inhibitory activity on the expression of a protein encoded by the target gene.
