<P>PROBLEM TO BE SOLVED: To provide a genetic method for tethering polypeptides to a yeast cell wall in a form accessible to protein-protein binding. Ž<P>SOLUTION: There is provided the method for selecting proteins on enhanced displayability exceeding a wild type protein on a yeast cell surface. The method includes the steps of: transforming yeast cells with a vector expressing a protein to be tested fused to a yeast cell wall protein, wherein mutagenesis is used to a generate a variegated population of mutants of the protein to be tested; bringing the yeast cell into contact with a label which binds to proteins that are displayed on the yeast cell surface and does not bind to proteins that are not displayed on the yeast cell surface; and the step of isolating the yeast cell to which the label binds, wherein the enhanced presence of the label bound to the protein to be tested indicates that the protein to be tested can be displayed on the yeast cell surface.
