<P>PROBLEM TO BE SOLVED: To obtain new rare-cutting endonucleases which have new sequence specificity about recognition and cleavage. <P>SOLUTION: There are provided hybrid and/or single-chain rare-cutting endonucleases, called meganucleases, which recognize and cleave a specific nucleotide sequence; polynucleotide sequences encoding the rare-cutting endonucleases; a vector comprising one of the polynucleotide sequences; a cell or non-human animal comprising one of the polynucleotide sequences or the rare-cutting endonucleases; and a method for producing one of the rare-cutting endonucleases and any use of the products and methods.
