摘要 PROBLEM TO BE SOLVED: To provide a method for quantitatively determining a nucleic acid molecule in a specimen in high sensitivity and accuracy. SOLUTION: The method for quantitative determination of a target nucleic acid molecule in a nucleic acid-containing specimen includes (a) the step to prepare a specimen solution by adding the nucleic acid-containing specimen, a first nucleic acid molecule probe having a base sequence complementary to the target nucleic acid molecule bonded with a first marker, and a second nucleic acid molecule probe having a base sequence complementary to the first nucleic acid molecule probe bonded with a second marker, (b) the step to denature the nucleic acid molecule in the specimen solution prepared by the step (a) followed by hybridization of the denatured molecule, and (c) the step to quantitatively determine the target nucleic acid molecule by detecting a temporal change of optical properties of the first marker or the second marker in the specimen solution after the step (b); wherein at least one of the first marker and the second marker is a substance changing its optical properties between the state that the first nucleic acid molecule probe is hybridized with the second nucleic acid molecule probe and the state that the first probe is not hybridized with the second probe.
