<P>PROBLEM TO BE SOLVED: To provide compositions and methods for amplification of nucleic acid sequences of interest. <P>SOLUTION: The method comprises incubating nucleic acids including the sequence of interest, at a high temperature, in the presence of heat-unstable nucleic acid polymerase having strand substituting activity, additives and a pair of primers and under conditions promoting replication of the nucleic acids, wherein the replication of the nucleic acid produces the replicated strands as the result, at least one of the replicated nucleic acid strands is substituted by strand substitution replication of another replicated strand, formation of the strands replicated from the sequences of interest is more advantageous than that of strands replicated from the sequences of no interest. <P>COPYRIGHT: (C)2010,JPO&INPIT |
