Provided is a method for preparing genetically engineered N-terminal acetylated thymosin a1, which includes the following steps: 1) preparing precursor proteins or polypeptides comprising N-terminal acetylated thymosin a1 polypeptide sequence by use of genetically engineered Escherichia coli; and 2) cleaving precursor proteins or polypeptides comprising N-terminal acetylated thymosin a1 polypeptide sequence of step 1) by endopeptidase, then purifying the resulting mixture to obtain N-terminal acetylated thymosin a1.
