PURPOSE: A plasmid vector for producing water-soluble light chain antibody fragment is provided to have Fab antibody library without contamination of helper phage. CONSTITUTION: A method for producing transformant of dual vector system-II-1(DVS-I-A) comprises: a step of binding light chain of human antibody to Pbr322 to obtain a first vector; a step of performing PCR of pBAD/gIII plasmid with primers of sequence numbers 3 and 4 to obtain a first DNA fragment; a step of performing PCR of pCDFDuet-1 plasmid with primer set of sequence numbers 5 and 6 to obtain a second DNA fragment; a step of binding the first and second DNA fragment and binding with the heavy chain of human antibody to obtain a second vector; a step of transforming the first vector with a host cell; and a step of transforming the second vector with the transformed host cell.
