PROBLEM TO BE SOLVED: To provide a soluble Spo11 protein holding DNA binding ability, and to provide a method for preparing the protein.

SOLUTION: The method for preparing the soluble Spo11 protein holding the activity includes simultaneously expressing the Spo11 protein and a trigger factor or a fused protein of the trigger factor and the Spo11 protein in a host Escherichia coli, crushing cells of the host Escherichia coli, collecting the soluble component, and preparing the soluble Spo11 protein from the soluble component.
