A matrix extracted from fibroblast of placenta is provided to show significant effect as a substrate required for adhering and spreading mesenchymal stem cells derived from cord blood and improve the expression of Oct-4. A substrate for propagation or adhesion of mesenchymal stem cells derived from cord blood includes matrix extracted from fibroblast of placenta. A method for preparing the substrate comprises the steps of: (a) culturing the fibroblast of the placenta in a culture medium on a plate; (b) removing the culture medium therefrom; (c) adding an extraction buffer solution thereto and culturing until cells are not seen; (d) washing cell remains; and (e) treating with DNase. Further, the method for preparing the substrate additionally comprises a step of adding antibiotic to the plate after treating with the DNase.
