An antioxidant composition comprising the enzymatic extract of Picrorrhiza kurroa is provided to remove DPPH(diphenylpicrylhydrazyl) radical, hydroxyl radical and alkyl radical, and protect DNA damaged by free radicals without side effects and toxicity, thereby inhibiting or treating aging and diseases caused by reactive oxygen-producing oxides. An antioxidant composition comprises the enzymatic extract of Picrorrhiza kurroa which is prepared by solubilizing Picrorrhiza kurroa in phosphate buffer or sodium acetate-acetic acid buffer with pH 3.0-8.0, adding a protein hydrolase selected from protamex, flavourzyme, neutrase, trypsin and alcalase or a sugar hydrolase selected from promozyme, celluclast, maltogenase, viscozyme, termamyl, dextrozyme and amyloglucosidase into the solution, stirring the mixture at 25-70 deg. C for 6-10 hours, heating the reacted mixture at 90-100 deg. C for 10 minutes, and concentrating the reacted solution under reduced pressure and freeze-drying it. The antioxidant composition is used as medicines, health supplement, cosmetic, food or food additive.
