A micro-channel chip for counting specific micro-particles among floating mixed micro-particles by an optical method and a method for counting micro-particles by using the same are provided to obtain a clear image without overlapping by setting up a depth of micro-channel to a depth of field in the micro-channel chip. A micro-channel chip for counting specific micro-particles among floating mixed micro-particles by an optical method includes a micro-channel to receive a sample including the micro-particles. The micro-channel chip counts the number of micro-particles in the micro-channel by taking photographs of inner parts of the micro-channel. The depth of the micro-channel is a depth of field to detect subjects clearly, wherein the depth of field is a range of focus.
