A light guide plate, a backlight unit, and a method of manufacturing the backlight unit are provided to obtain high brightness and implement a mass production by forming an optical pattern with a V groove and a protrusion portion integrated with the V-groove. A light guide plate includes a main body(323) and an optical pattern portion(322). The main body receives light which is emitted from a lamp. The optical pattern portion is formed on one surface of the main body and normally reflects the incident light. The optical pattern portion includes a cutting surface and a protrusion portion. The cutting surface has a V-groove. The protrusion portions are formed at both sides of a cutting surface. When the light guide plate is moved along an inline, a cutter on the inline is line-contacted with one surface of the light guide plate, so that one surface of the light guide plate is softened. During the softening process, the cutter scratches the one surface to form the V-groove and the protrusion portion. The V-groove and the protrusion portion are cured.
