PROBLEM TO BE SOLVED: To provide a highly-sensitive target substance detection material, and its preparation method. SOLUTION: The target substance detection material for detecting a target substance in specimen liquid by utilizing a plasmon resonance method is a particulate multilayer structure having a particulate center body having a low refractive-index material and a metal including layer coating the outer circumference of the center body. The preparation method of the target substance detection material for detecting the target substance in the specimen liquid by utilizing the plasmon resonance method is a preparation method of the particulate multilayer structure having a process for preparing liposome including water inside, a process for carrying metal particles on the liposome surface, a process for forming an outer layer by growing a metal including film by using each metal particle as a nucleus, and a process for carrying a capturing body component for capturing the target substance in the specimen liquid on the outer layer surface.
