A recombinant microorganism, which is an yeast two hybrid system, is provided to identify the interaction between proteins through a simple process since a separate substrate is not required for expressing a reporter gene. A recombinant microorganism comprises a DNA encoding an N-terminal domain of split enhanced green fluorescent protein(NEGFP) of EGFP(enhanced green fluorescent protein), which has a CNE6/ARS4 origin and includes a sequence of SEQ ID : NO. 1, and a DNA encoding a C-terminal domain of split enhanced green fluorescent protein(CEGFP) of the EGFP and is transformed by a recombinant vector in which a GAL4DD(GAL4 dimerization domain) is conjugated into a 3'-terminal of the NEGFP encoding DNA as a bait polynucleotide and a GAL11P(GAL11 protein) is conjugated into a 3'-terminal of the CEGFP encoding DNA as a prey polynucleotide. A method for detecting the interaction between a bait and a prey comprises a step of culturing the recombinant microorganism and then measuring the fluorescence of the cultured recombinant microorganism.
