摘要 A new cancer stem cell line GBM 2 established from human Glioblastoma multiforme tissue is provided to identify a cancer stem cell as a target for cancer treatment, so that the cancer stem cell is useful for screening anticancer agents and developing therapy for maximizing cancer treatment efficiency. A new cancer stem cell line GBM 2(KCLRF-BP00134) is derived from the Glioblastoma multiforme tissue of malignant brain tumor patients, and has drug resistance against anticancer agents and transdifferentiation into nerve cells after expression and dedifferentiation of nestin and Oct 4 from undifferentiated nerve stem cells. A method for preparing the cancer stem cell line GBM 2(KCLRF-BP00134) comprises the steps of: collecting the Glioblastoma multiforme tissue from a malignant brain tumor patient; cutting the tissue and treating the cuts with trypsin to separate cells; adding bFGF(basic fibroblast growth factor) and EGF(epidermal growth factor) into the separated cells and culturing them; subculturing the cells to establish the cancer cell line; identifying a subpopulation positive to CD133 from the cancer cell line through FACS(vantage fluorescence-activated cell sorter); confirming expression of nestin and Oct 4 through RT-PCR(reverse transcription-polymerase chain reaction) of the cancer cell line; dedifferentiating the cancer cell line and confirming the transdifferentiation into nerve cells; and confirming the drug resistance against anticancer agent of the cancer cell line.
