An ultrasound system for controlling a sample volume and a controlling method thereof are provided to measure a blood flow rate by accurately aligning a position of the sample volume at a center of a blood vessel. An image signal acquiring unit acquires first and second image signals. An image forming unit forms a B-mode ultrasound image based on the first image signal and a Doppler spectrum image based on the second image signal. A display unit displays the B-mode ultrasound image and the Doppler spectrum image. An input unit receives set information for a sample volume and set information for a scan region for detecting a maximum speed of a blood flow. A sample volume and scan region setting unit sets the sample volume on the B-mode ultrasound image based on the sample volume set information and sets the scan region based on the scan region set information and the sample volume. A sample volume processor(170) controls the sample volume.
