摘要 <p>Identification of an active substance complex that modifies an enzymatic characteristic of a target molecule having a palmitoyl acyl transferase 1 (PAT1) type of protein, comprises: (a) providing a number of primary carriers containing the target molecules; (b) contacting the primary carriers within a bio-sensor electrode; (c) providing at least a potential complex of an active substance; (d) contacting the potential complex with the target molecule; and (e) determining the qualitative or quantitative influence of the potential complex on enzymatic characteristics of the target molecule. Identification of an active substance complex that modifies an enzymatic characteristic of a target molecule containing a palmitoyl acyl transferase 1 (PAT1) type of protein, comprises (a) providing a number of primary carriers that contains a number of the target molecules containing the PAT1 type of protein within the membrane region of the respective primary carrier; (b) superpositioning or bringing the primary carriers in contact at or within the surface area of an isolation region of a bio-sensor electrode serving as a secondary carrier in a measuring medium, where the secondary carrier is mechanically and electrically isolated with respect to the measuring medium and to the primary carriers by the isolation region; (c) providing at least a potential complex of an active substance; (d) bringing the potential complex in contact with the target molecule of the primary carrier or parts of it; and (e) determining the qualitative or quantitative influence of the potential complex or a part of it on enzymatic characteristics of the target molecule or a part of it through detection of an electrical action of the target molecule or a part of it or changing the electrical action with the help of the bio-sensor electrode, where at least one of the process steps (c), (d) and (e) consist of one or more of the following sub-steps that are: (f) inserting the secondary carrier with the primary carriers into a quiescent medium and detecting an electrical action as in step (e); (g) inserting the secondary carrier with the primary carriers into a second non-activating medium and detecting an electrical action as in step (e); (h) inserting the secondary carrier with the primary carriers into a third or activating measuring medium and detecting an electrical action as in step (e); where the third activating medium corresponds to the second non-activating medium, additionally contains a substrate or a co-substrate of the PAT1 type of protein or a concentration of a substrate or co-substrate of the PAT1 type of protein that is varied with respect to the non-activating medium, where a medium with 100 mmol/l potassium chloride, 2 mmol/l magnesium chloride, 30 mmol/l Mes pH 6/potasssium hydroxide and valine within the range of approximately 2 mmol/l to approximately 80 mmol/l is used as non-activating medium and where a medium with 100 mmol/l potassium chloride, 2 mmol/l magnesium chloride, 30 mmol/l Mes pH 6/potassium hydroxide and glycine within the range of about 2 mmol/l to approximately 80 mmol/l is used as activating medium. Independent claims are included for: (1) a test kit for the execution of the above process comprising: at least a primary carrier; a measuring region; a first quiescent medium; a second non-activating medium and a third activating medium; and (2) a screening process for identifying an unknown active substance, complex of an active substance, a part or a derivative of it; the presence of an unknown active substance, a complex of an active substance, a part or derivative of it; the presence of a well-known active substance, a complex of an active substance, a part or derivative of it; the concentration of an unknown active substance, a complex of an active substance, a part or derivative of it; the concentration of a well-known active substance, a complex of an active substance, a part or derivative of it; or any combination specified of the previously mentioned parameters by using the above method or the test kit, where the active substance, the complex of an active substance, the part or the derivative of it is modified by an enzymatic characteristic of a target molecule or a part of it containing a PAT1 type of protein, or is transferred by PAT1.
