PROBLEM TO BE SOLVED: To provide a pseudo moving layer type chromatographic separation method capable of acquiring simply a high separation performance, concerning a chromatographic separation method using an adsorbent for optical resolution as an adsorbent. SOLUTION: In this chromatographic separation method using a chromatographic column filled with the adsorbent for optical resolution, which is the pseudo moving layer type chromatographic separation method for refining by chromatographic separation, separation is executed by combining the first process wherein at least either of undiluted solution and eluate is supplied into a circulation system, and a quantity equal to a liquid quantity supplied into the circulation system is drawn out from one spot of a packed tower wherein some component is enriched in the circulation system, and the second step wherein the liquid in the system is circulated without supplying the undiluted solution and the eluate at all and without drawing out a separated component.
