Novel N-substituted 4-butoxy-2,6-dimethylbenzamides of the formula wherein X is CH 2 or a direct link and the non- toxic acid addition salts thereof are prepared either by reacting 1-methyl-2(or 3- or 4-)- aminomethylpiperidine or 1 -methyl-2(or 3-)- aminomethylpyrrolidine with 4-butoxy-2,6-dimethylbenzoic acid or a reactive functional derivative thereof or by reacting the appropriate aminomethylpyridine or aminomethylpyrrole with 4-butoxy-2,6-dimethylbenzoic acid or reactive functional derivative thereof, hydrogenating and methylating the thus obtained amide, the said methylation being carried out either before or after hydrogenation. 1 - Methyl - 2 - aminomethylpiperidine is obtained by hydrogenating N-methylpyridinium- 2-aldoxime iodide. 2 - (and 3 -) - (41 - Butoxy - 2<SP>1</SP>,6<SP>1</SP> - dimethylbenzoylamino)-methylpyridine is prepared by reacting 2-(or 3-)aminomethylpyridine with 4- butoxy-2,6-dimethylbenzoyl chloride. Pharmaceutical compositions, having local anaesthetic activity and suitable for parenteral administration contain the above novel compounds or non-toxic acid addition salts as the active ingredient.</p> |
