PROBLEM TO BE SOLVED: To provide a highly sensitive and simple method of measuring membrane potential change utilizing a membrane potential sensitive fluorescent pigment. SOLUTION: The method of measuring the membrane potential change has a step for acquiring a pigment-introduced cell by executing introduction of a fluorescence quenching pigment into an intracellular or extracellular medium and staining of a cell membrane of a cell by the membrane potential sensitive fluorescent pigment, simultaneously or in the order wherein either is executed in advance; and a step for measuring with the passage of time the fluorescence intensity generated from the membrane potential sensitive fluorescent pigment by irradiating the pigment-introduced cell with excitation light.
