1357315 Enzyme-cellulose complex; polynucleotides MERCK & CO Inc 3 May 1971 [4 May 1970] 59095/73 Divided out of 1355251 Headings C3H and C3A An enzyme is brought together with cyanogenhalide-activated cellulose in an aqueous medium, whereupon the enzyme becomes covalently bound to the cellulose. The cellulose may first be mercerized with alkali (e.g. NaOH), then washed and centrifuged. The resultant slurry is treated at 0‹ C. with an aqueous solution of cyanogen bromide plus enough NaOH to give a pH of 10 to 11. The product is washed with water, then with a buffer solution and combined with an aqueous solution of polynucleotide phosphorylase at 5‹ C. under nitrogen for 15 hours; the product is centrifuged and washed. Inosine diphosphate treated at 37‹ C. with the above insolubilized enzyme in the presence of unfractionated oligoadenylic acid and trisbuffer at pH 8À2 gives a solution of polyinosinic acid from which the enzyme may be removed by centrifugation and re-used. Polycytidylic acid may similarly be obtained. |
