PROBLEM TO BE SOLVED: To provide a sample measuring device and a measuring method capable of measuring properly a sample such as a cell. SOLUTION: This device is provided with a sample holding mechanism 10 for holding a liquid L including the sample S such as a cell which is a measuring object; and a phase difference image acquiring device 20 for irradiating a measuring domain R in the sample holding mechanism 10 with measuring light from a measuring light source 21 along three mutually different measuring directions D1-D3, measuring light passing through the sample S in each measuring direction D1-D3 by an imaging device 22, and acquiring its phase difference image. A prescribed analysis is performed by an image reconstitution device 30 relative to three phase difference images in each measuring direction D1-D3 acquired by the phase difference image acquiring device 20, and the image of the sample S in the measuring domain R in the sample holding mechanism 10 is reconstituted based on the analysis result.
