摘要 PROBLEM TO BE SOLVED: To create/provide an Arabidopsis thaliana-derived minichromosome-holding strain usable for chromosome function analysis and artificial chromosome development and holding a new minichromosome differing in origin/properties from known minichromosomes. SOLUTION: The new minichromosome is developed when a marker gene-integrated vector is transferred into a strain Tr4SCo5 holding a minichromosome 4S derived from the short arm of Arabidopsis thaliana No.4 chromosome. As the result of its analysis, it has been clarified that this new minichromosome 2S is derived from the short arm of the No.2 chromosome. Because this minichromosome 2S contains GFP gene and kanamycin-resistant gene, the minichromosome-holding strain can be selected by kanamycin and emits green fluorescence. Further, when two of this minichromosome transmit to the later generations, the phenomena that both kanamycin resistance and green fluorescence vanish are observed. COPYRIGHT: (C)2006,JPO&NCIPI
