<P>PROBLEM TO BE SOLVED: To provide a DNA amplifying method capable of simply and efficiently preparing single-strand DNA in a desired region in a desired direction. <P>SOLUTION: The DNA amplifying method comprises using DNA as a template. The amplifying method comprises a first process for carrying out PCR cycle at an annealing temperature Ta (wherein Ta ≤Tm<SB>1</SB>and Ta ≤Tm<SB>2</SB>) by using a first primer which is complementary to 3' end side of a target base sequence, a second primer having a base sequence which is homogeneous to 5' end side of the target base sequence and a third primer having a base sequence which is homogeneous to a partial base sequence on 5' end side of the second primer or the first primer on the 3' end side under conditions in which melting temperature Tm<SB>1</SB>of the first primer, a melting temperature Tm<SB>2</SB>of the second primer and melting temperature tm<SB>3</SB>of the third primer have relationships represented by formulas Tm<SB>1</SB><Tm<SB>3</SB>and Tm<SB>2</SB><Tm<SB>3</SB>and a second process for carrying out PCR cycle at an annealing temperature Tb (wherein Tm<SB>1</SB><Tb and Tm<SB>2</SB><Tb and Tb≤Tm<SB>3</SB>). <P>COPYRIGHT: (C)2006,JPO&NCIPI |
