PROBLEM TO BE SOLVED: To provide a method for detecting microorganisms involving methanation existing in methane fermentation treatment sludge of organic waste and waste water and to provide a primer used for the method.

SOLUTION: The present invention provides (a) a primer corresponding to a base sequence encoding 16SrRNA which is common to whole bacteria involving methane fermentation or (b) a primer specific to the genus Clostridium or (c) a primer specific to the genus Methanothermobacter.
