The invention provides methods of expressing an siRNA starting from a single template encoding one strand of the siRNA. The methods are particularly suitable for generating large libraries of random siRNAs in which one strand of each siRNA can be synthesized simultaneously in a solid-phase synthesis procedure. Methods for normalizing siRNA libraries to enrich for siRNAs of interest and methods of expressing siRNAs are also provided.
