A multi-chamber electrode assembly for electrophysiological recording comprising a chamber plate 106 with a plurality of extracellular compartments 108, a partition plate 86 with a plurality of electrode apertures 18 aligned with said extracellular compartments 108, spacers 112 between said chamber plate 106 and partition plate 86 to form fluidic channels 122 for withdrawing fluid therethrough from the extracellular compartments 108, a foundation plate 88 with a plurality of intracellular compartments 90 aligned with said electrode apertures 18. The intracellular compartments 90 each include an electrode 92 connectable to electrophysiological recording equipment. The foundation plate 88 further comprises fluid-flow conduits aligned with said fluidic channels 22 for withdrawing fluid from the extracellular compartments 108. The assembly is made by stacking the foundation plate 88, partition plate 86, spacers 122 and chamber plate 108.
