PROBLEM TO BE SOLVED: To provide a method by which a DNA library in or from which a desired DNA is specifically concentrated or removed can be produced from a DNA library to directly obtain the clone of the DNA.

SOLUTION: The method for constructing a cyclic DNA library containing the desired DNA in an increased content comprises removing a specific DNA from a cyclic DNA library with RecA protein. Namely, the method for constructing the cyclic DNA library containing the first dsDNA in the increased content comprises removing a second DNA different from the first dsDNA from the DNA library containing the dsDNA to be increased.
