PROBLEM TO BE SOLVED: To provide a C terminal amino acid sequence analysis method of a peptide utilizing a decomposition reaction means of a sequential C terminal amino acid, capable of suppressing an undesirable side reaction such as cut of a peptide bond at the middle of the peptide when decomposing sequentially the C terminal amino acid of the peptide having the long amino acid length, and executing the processing in a versatile condition. SOLUTION: N-acylation is applied beforehand to a dry sample of the peptide having the long amino acid length, and sequential decomposition of the C terminal amino acid is performed in a mild condition by utilizing a reaction reagent formed by combining an alkanoic acid anhydride with a small quantity of perfluoro alkanoic acid. After hydro-processing, fragmentation is performed selectively at an arginine residue portion by trypsin digestion, and molecular weight reduction of a C terminal fragment derived from a series of reaction products is measured by negative mode analysis of a MALDI-TOP-MS device, to thereby specify the C terminal amino acid sequence.
