PURPOSE: Provided is a biological soil remediation method using liquid microbial products capable of rapidly cultivating microorganisms which can degrade contaminant in soil water purification method and acceleration nutrient capable of activating the microorganisms. CONSTITUTION: The method comprises the steps of in-situ producing microbial products by rapidly cultivating microorganisms at the spot and introducing activation acceleration nutrient and the microbial products into contaminated soil, wherein the producing microbial products includes (a) sampling 100 to 500g of soil deep in depth of 0 to 30 cm from the surface of earth in contaminated area and peripheral area of the contaminated area, (b) preparing culture medium by adding 2 to 7g of aluminum chloride, 1 to 3g of sodium phosphate 1 to 3g, 0.2 to 0.6g of EDTA, 0.3 to 0.5g of potassium chloride, 0.2 to 0.4 sodium sulphate, 0.2 to 0.4g of magnesium sulphate and 5 to 10 mL of mineral solution to 1L of distilled water, (c) feeding 5 to 50g of the sampled soil to 100 mL of the culture medium, suspension-culturing it for 10 to 14 hours, settling the solution for 1 hour and taking 50 to 100 mL of decant water of the solution, (d) feeding carbon source to the decant water by 100 to 10000 ppm, culturing the mixture at 25 to 35°C and 100 to 200 rpm for a week, and taking 10 mL of the cultured matter, (e) repeating the step (d), (f) diluting the cultured matter with a dilution ratio of 100 to 10000 times, smearing the diluted matter onto a sterilized agar medium, and culturing it for a week, (f) separating colonies on the agar medium, pure-culturing the colonies, performing centrifugal separation, free drying the separated microbes and reserving the microbes at -60 to -80°C, (g) suspension-culturing microbe strains obtained at the sep (f) using a culture medium which is prepared by adding 5 to 10g/L of glucose to the culture medium obtained at the step (b) for two days. |
