PURPOSE: An inhibitor of protein kinase is provided to prevent and treat protein kinase-related abnormal conditions in organisms, by using partly phenol- and hydroxynaphthalene-based compounds. CONSTITUTION: The inhibitor of protein kinase comprises a compound of formula 1 or a pharmaceutically acceptable salt, amide, ester, or prodrug thereof, wherein (a) R1 is selected from the group consisting of (i) a six-membered aromatic or heteroaromatic, or a five- or six-membered heteroaromatic ring, optionally substituted with one or more substituents, (ii) an acyl having a structure of -(X1)n1-C(O)-X2; and (iii) a substituent having a structure of -C(X4)=N-NX5-C(=E)-NX6X7, or of -C(X4)=N-N=C(EX5)-NX6X7; (b) R2, R3 and R4 are each independently selected from the group consisting of (i) hydrogen, (ii) lower alkyl, (iii) lower alkylene, (iv) halogen or perhaloalkyl, (v) an alkoxy having a structure of -(X13)n13-O-X14, and (vi) a five-membered or six-membered heteroaryl ring or a six-membered aryl or heteroaryl ring; (c) R5 is selected from the group consisting of hydrogen, lower alkyl, aryl, and heteroaryl; (d) R100 is selected from the group consisting of hydrogen, lower alkyl, lower alkene, lower alkyn, aryl, and heteroaryl; and (e) E1 is selected from the group consisting of hydrogen, sulfur, and -NR101-, provided that at least one of R1-R5 is not selected from the group consisting of hydrogen, lower alkyl, aryl, and heteroaryl.
