PROBLEM TO BE SOLVED: To provide a multilayer liposome which is readily prepared and is stable with time and a multilayer liposome preparation. SOLUTION: The multilayer liposome comprises (A) a polar lipid, (B) a nonionic surfactant having 1-9 HLB, (C) a nonionic surfactant having >9 and≤20 HLB and water in a weight ratio [(C)/(A) + (B)] of 0.8-3.5. The multilayer liposome preparation is obtained by formulating the multilayer liposome with (E) a physiologically active substance. The multilayer liposome which is readily prepared and is stable with time and has a high concentration can simply and stability be mass-produced. The multilayer liposome can be used for applications e.g. cosmetics, external preparations such as medicines, fertilizers and agrochemicals as a highly functional material, because a water-soluble, an oil-soluble or an amphipathic physiologically active substance can be encapsulated in the multilayer liposome.
